Abstract: The increasing threat of antimicrobial resistance (AMR) has intensified the search for new bioactive compounds from natural sources. In this context, stingless bees and their products represent promising alternatives due to their rich microbial symbioses. This study investigated the antimicrobial activity of honeys produced by Scaptotrigona bipunctata, a stingless bee species endemic to Brazil, and aimed to isolate microorganisms potentially responsible for this activity. Honey samples were collected from five different localities in Toledo, Paraná, Brazil. Antimicrobial susceptibility was evaluated against Staphylococcus aureus, Escherichia coli, Bacillus cereus, and Streptococcus pyogenes using agar well diffusion assays. All samples inhibited S. aureus, while no activity was observed against E. coli. The most active sample also inhibited B. cereus and S. pyogenes. A total of 25 bacterial isolates were obtained from the honeys under aerobic and anaerobic conditions. Two isolates exhibited antagonistic activity against S. aureus, with their cell-free supernatants (CFS) producing inhibition zones and showing minimum inhibitory concentrations (MICs) of 25%. The isolates were initially identified as Gram-positive bacteria. These findings indicate that S. bipunctata honey and its associated microbiota are potential sources of antimicrobial compounds, particularly against Gram-positive bacteria, underscoring their biotechnological relevance for the development of novel therapeutic agents.

Abstract: The present work aimed to develop electrospun nanofibers based on NPK with the polymers Ecovio®, polyvinyl alcohol (PVA) and cationic starch, aiming at the production of a material with modified release of nutrients. The material obtained was of the Ecovio/(PVA/Starch)/Ecovio sandwich type. The fibers were obtained by electrospinning, and characterized by various physical, chemical and mechanical techniques. The incorporation of NPK significantly increased the conductivity of the solutions (from 0.628 mScm-1 to 9.66 mScm-1 in PVA/Starch), favoring electroreliability. The fibers with nutrients (F1) had a lower amount of beads and an average diameter of 503.86 ± 8.4 nm, compared to 516.45 ± 15.04 nm of the reference (R1). The mechanical analysis showed an increase in the breaking stress (0.71 MPa vs. 0.36 MPa), elongation (41.02% vs. 30.8%) and Young's modulus (6.06 MPa vs. 2.02 MPa). The conductivity of the water indicated rapid release of nutrients, with a peak of 16.19 μScm-1 in 24 hours. TOC analysis revealed release of up to 5.43 mgL-1 of organic carbon and 1.47 mgL-1 of nitrogen in 28 days. The lettuce seed germination assay demonstrated similar performance to the conventional method, indicating potential for agricultural application. Despite the promising results, improvements in the hydrophobicity of the material are necessary to prolong the release of nutrients.

Abstract: Psoriasis is one of the most common skin diseases in the world. Its main clinical manifestation is the presence of erythematous and pruritic plaques that affect approximately 90% of cases. Treatment of lesions results in an improvement in the quality of life of patients and is carried out mainly with the use of topical corticosteroids. As an alternative to conventional treatment of psoriasis, this work presents a study on the production and characterization of polymeric membranes containing betamethasone valerate produced by biaxial electrospinning. The selected polymers were PVP and ECOVIO®. In addition to being biocompatible and stable at pH and temperature, PVP has high solubility in water, resulting in rapid release of the incorporated drug. ECOVIO®, composed of PLA and PBAT polymers, has good mechanical properties and forms membranes with controlled drug release, due to its hydrophobic characteristics. The produced membranes were characterized by SEM, FTIR, TGA, DSC and XRD techniques. Good compatibility between the membrane components was observed, as well as increased drug solubility, observed in the in vitro release study. Additionally, PVP showed a rapid release profile, releasing approximately 90% of the active ingredient in 5 minutes, while the release from the ECOVIO® membrane occurred in a controlled manner over 24 hours. For the biaxial membrane, a faster initial release was observed in relation to that obtained in the ECOVIO® membrane, referring to the PVP portion of the membrane, followed by a controlled release for 24 hours, characteristic of the ECOVIO® portion. Thus, according to the results obtained, it can be concluded that the membrane produced is promising for the controlled release of betamethasone valerate.

Abstract: The U.S. Department of Health, through their ICH guides, describes Quality by Design (QbD) as "a systematic approach to pharmaceutical development that begins with predefined objectives and emphasizes product, process understanding and control, based on sound scientific data and quality risk management." The concept of QbD can and should be applied to analytical methods, since they are an integral part of pharmaceutical development. Thus, arises Analytical Quality by Design (AQbD), a justifiable and recommended procedure to reduce out-of-specification results, achieve a high degree of robustness, and a cost-effective analytical method and regulatory flexibility. Design of Experiments (DoE) is an important tool set that includes screening and optimization designs to study variation in factors and determine their effects on responses, thus identifying which factors are most significant. This work aimed to apply the concepts of AQbD tools in the development and validation of an analytical method for the residual solvents acetone, methanol, dichloromethane and chloroform in the raw material Bisoprolol Hemifumarate by gas chromatography with flame ionization detection (FID) and headspace transferline (HS) injection mode. In the first step of the study, a factor screening was performed using the Plackett-Burman design to define the best response conditions with maximization of signals and recovery of analytes. With the variables filling pressure and incubation temperature being the most significant, the central composite design (CCD) was used as an optimization experiment in 5 levels and 11 trials, 4 in cube, 4 in axial and 3 central points, with α=√2 where, at the end, a tendency to better results was obtained. A robustness study, a parameter typically performed in method development, followed with the best condition of the parameters fill pressure and incubation temperature, in addition to others typically used in robustness. Then, the analytical validation step was performed according to RDC 166/2017 demonstrating that the method is selective, linear, precise and accurate method. The application of the AQbD tools allowed a better understanding of the variables, leading to the development of an analytical method by gas chromatography in a rational way and achieving all the criteria required.
